A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Benralizumab Antibody, clone AbD59979oap (TZA097P) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with benralizumab, IL5Ra-His, and human IgG1 kappa control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, the IL5Ra-His/benralizumab complex was formed by adding 5 µg/mL benralizumab to the wells coated with 5 µg/mL IL5Ra-His. Detection was performed using Human Anti-Benralizumab (Drug/Target Complex) Antibody, clone AbD59979ad (TZA097) titrated to the given concentrations followed by an HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for IL5Ra-His/benralizumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of benralizumab. Detection was performed using HRP-conjugated Human Anti-Benralizumab (Drug/Target Complex) Antibody, clone AbD59979oap (TZA097P) at a concentration of 0.2 µg/mL in HISPEC Assay Diluent (BUF049A), and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Benralizumab (Drug/Target Complex) Antibody, clone AbD59979 specifically recognizes the antibody drug benralizumab when in complex with its target, human interleukin 5 receptor α (IL5Rα ). It does not recognize free benralizumab nor unbound IL-5R. The monovalent Fab format of this antibody, clone AbD59979ad (TZA097) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a monovalent Fab antibody pre-coupled to the HRP-conjugated SpyCatcher2 (TZC002P) under product code TZA097P for direct detection of the benralizumab/ IL5Rα complex in bioanalysis assays. Clone AbD59979 can be used in pharmacokinetic (PK) antigen detection format assays to measure benralizumab that has been captured via immobilized IL5Rα. Benralizumab (trade name Fasenra) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds with high affinity to a conformationally distinct epitope within domain 1 of the α-chain of IL-5R, and thus inhibits proliferation of IL-5-dependent cell lines, and otherwise blocks signaling through the IL-5 receptor. - A monovalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA097 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
