A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Benralizumab Antibody, clone AbD59973pap (TZA069P) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with benralizumab, IL5Ra-His, human IgG1 kappa control and normal human serum (NHS) at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Benralizumab Antibody, clone AbD59973ad (TZA069) titrated to the given concentrations followed by an HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for benralizumab are shown as the mean of three measurements.
A microtiter plate was coated overnight with Human Anti-Benralizumab Antibody, clone AbD60095ad (TZA068) at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of benralizumab. Detection was performed using HRP-conjugated Human Anti-Benralizumab Antibody, clone AbD59973pap (TZA069P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL5Ra-His at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of benralizumab (0.15 µg/mL) plus increasing concentrations of Human Anti-Benralizumab Antibody, clone AbD59973ad (TZA069). Free benralizumab, still capable of binding to the IL5Ra-His coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Benralizumab Antibody, clone AbD59973 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug benralizumab. It does not recognize the drug target, human interleukin 5 receptor α (IL5Rα), nor benralizumab in complex with IL5Rα. This antibody can be used to measure free benralizumab levels in serum from patients. Clone AbD59973 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA069. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA069P. A pair of anti-benralizumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD59973ad is recommended as a detection antibody, paired with Human Anti-Benralizumab Antibody, clone AbD60095ad (TZA068) as the capture reagent. Benralizumab (trade name Fasenra) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds with high affinity to a conformationally distinct epitope within domain 1 of the α-chain of IL-5R, and thus inhibits proliferation of IL-5-dependent cell lines, and otherwise blocks signaling through the IL-5 receptor.
