A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Elotuzumab Antibody, clone AbD60184pap (TZA0116P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with elotuzumab, human VH/VL subclass control, Rituximab, Alemtuzumab and Trastuzumab at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Elotuzumab Antibody, clone AbD60184pap (TZA0116P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for elotuzumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of elotuzumab. Detection was performed using HRP-conjugated Human Anti-Elotuzumab Antibody, clone AbD60184pap (TZA0116P) at a concentration of 2 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, the plate was coated with human SLAM7-His-bio at a concentration of 1 µg/mL and washed. A pre-incubated mixture of Elotuzumab (0.15 µg/mL) plus increasing concentrations of Human Anti-Elotuzumab Antibody, clone AbD60184ad (TZA0116) was added. Free elotuzumab, still capable of binding to the human SLAM7 coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. E Reset| Human Anti-Elotuzumab Antibody, clone AbD60184 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug elotuzumab, and its biosimilar products. It does not recognize the drug target, signalling lymphocytic activation molecule family member 7 protein (SLAMF7), nor elotuzumab in complex with SLAMF7. This antibody can be used to measure free elotuzumab levels in serum from patients. Clone AbD60184 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0116. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0116P. A pair of anti-elotuzumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD60184 is recommended as a detection antibody, paired with Human Anti-Elotuzumab Antibody, clone AbD60184ad (TZA0115) as the capture reagent. Elotuzumab (trade name Empliciti) is a humanized monoclonal antibody of the IgG1/kappa isotype that targets SLAMF7 and directly activates Natural Killer cells through both the SLAMF7 pathway and immunostimulatory Fc receptor binding. Elotuzumab also targets SLAMF7 on myeloma cells and facilitates the interaction with Natural Killer cells to mediate the killing of myeloma cells through antibody-dependent cellular cytotoxicity (ADCC). Elotuzumab is indicated to treat multiple myeloma and in combination with lenalidomide and dexamethasone for the treatment of patients with multiple myeloma. - A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA0116 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
