Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Romosozumab Antibody, clone AbD60148ia (HCA424) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Romosozumab Antibody, clone AbD60148ia (HCA424) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for romosozumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Romosozumab Antibody, clone AbD60141ia (HCA423, black), or clone AbD60148ia (HCA424, blue). Detection was performed using HRP conjugated romosozumab at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of romosozumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Romosozumab Antibody, clone AbD60148ia (HCA424) in the monovalent Fab format was added. Free Romosozumab, still capable of binding to the Sclerostin-His coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Romosozumab Antibody, clone AbD60148ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug romosozumab, and its biosimilar products. It does not recognize the drug target, sclerostin, nor romosozumab in complex with sclerostin. HCA424 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 2nM and is one of the two antibodies offered for ADA assays. Antibody HCA423 has an affinity of 0.7nM. Romosozumab (trade name Evenity) is a humanized monoclonal antibody of the IgG2/kappa isotype that binds to sclerostin and prevents inhibition of the Wnt signaling pathway, thereby promoting osteogenesis and, to a lesser extent, reducing bone resorption. Romosozumab's inhibition of sclerostin also inhibits the increase in RANKL-dependent increases in osteoclast activity and bone resorption.
