Mouse SeroBlock FcR is a rat monoclonal antibody (clone FCR 4G8) that specifically recognises mouse CD16 and CD32, which are cell surface proteins also known as FcRgIII and FcRgII respectively.
These proteins bind IgG molecules via their Fc regions as part of the adaptive immune response. CD16 and/or CD32 are expressed by a wide variety of cells, including monocytes, macrophages, B lymphocytes, granulocytes, NK cells, dendritic cells and some activated T lymphocytes. The expression of CD16/CD32 antigens can lead to non-specific binding of test monoclonal antibodies in staining procedures, leading to ‘high background’ staining on a wide range of cells. This non-specific binding may be blocked with SeroBlock FcR, resulting in clearer staining (see Figure 1).
SeroBlock FcR is fast acting, only requiring a 10 minute pre-incubation with target cells. It doesn’t interfere with specific antibody staining, and is suitable for both direct and indirect immunostaining.
Compared to an alternative clone available that also blocks Fc receptors, FCR 4G8 matches its high performance but is less expensive, meaning better value for money without a compromise in quality (see Figure 2).
Clone FCR 4G8 is also available for direct analysis of CD16/CD32 expression in unlabelled or conjugated antibody formats (see table). For functional studies of target cells where blockade of CD16/CD32 is desirable, the low endotoxin version (MCA2305EL) is recommended.
