Human Anti-Risankizumab Antibody, clone AbD59012ad is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug risankizumab, and its biosimilar products. It does not recognize the drug target, human Interleukin 23 (IL-23), nor risankizumab in complex with IL-23. This antibody can be used to measure free risankizumab levels in serum from patients. A pair of anti-risankizumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD59012ad is recommended as a capture antibody, paired with Human Anti-Risankizumab Antibody, clone AbD58231 (TZA059) as the detection antibody. TZA058 a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog. It is also available in the full length recombinant human IgG1 format under product code HCA414. Risankizumab (trade name Skyrizi) is a humanized monoclonal antibody of the IgG1/kappa isotype that suppresses the inflammatory effects of interleukin-23 by selectively targeting its unique p19 subunit, subsequently inhibiting IL-23 binding to its receptor and the signaling responsible for the release of pro-inflammatory cytokines and chemokines. Risankizumab is indicated as an interleukin-23 antagonist in the treatment of plaque psoriasis, psoriatic arthritis and Crohn's disease in adults. |
- Product Form
- A monovalent human recombinant Fab (kappa light chain) selected from the HuCAL phage display library and expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

- Technical Advice
- Recommended protocols for coupling a Fab antibody with a SpyTag to a SpyCatcher can be found at SpyTag and SpyCatcher Products, and further information about HuCAL recombinant antibody technology can be found in the HuCAL Antibodies Technical Manual.
- ELISA
- Clone AbD59012ad can be used in indirect ELISA to detect risankizumab. It can also be used as the capture antibody to develop a pharmacokinetic (PK) bridging ELISA for risankizumab, together with TZA059 or TZA059P as the detection reagent.
