A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Risankizumab Antibody, clone AbD58231papd (TZA059P) at a concentration of 0.2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Risankizumab, clone AbD58231ad (TZA059) titrated to the given concentrations followed by an HRP conjugated Anti-His Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for risankizumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of risankizumab. Detection was performed using HRP-conjugated Human Anti-Risankizumab Antibody, clone AbD58231pap (TZA059P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of risankizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Risankizumab Antibody, clone AbD58231ad (TZA059). Free risankizumab, still capable of binding to the IL-23 coated plate, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

| Human Anti-Risankizumab Antibody, clone AbD58231 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug risankizumab, and its biosimilar products. It does not recognize the drug target, human Interleukin 23 (IL-23), nor risankizumab in complex with IL-23. This antibody can be used to measure free risankizumab levels in serum from patients. Clone AbD58231 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA059. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA059P. A pair of anti-risankizumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD58231 is recommended as a detection antibody, paired with Human Anti-Risankizumab Antibody, clone AbD59012ad (TZA058) as the capture reagent. Risankizumab (trade name Skyrizi) is a humanized monoclonal antibody of the IgG1/kappa isotype that suppresses the inflammatory effects of interleukin-23 by selectively targeting its unique p19 subunit, subsequently inhibiting IL-23 binding to its receptor and the signaling responsible for the release of pro-inflammatory cytokines and chemokines. Risankizumab is indicated as an interleukin-23 antagonist in the treatment of plaque psoriasis, psoriatic arthritis and Crohn's disease in adults. |

- A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA059 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites.
