A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Nirsevimab Antibody, clone AbD64624ad (TZA0148) at a concentration of 2 μg/mL followed by HRP conjugated Anti-DYKDDDDK Tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with Nirsevimab, human IgG1 kappa control, human VH/VL subclass control, and normal human serum (NHS) at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Nirsevimab Antibody, clone AbD64624ad (TZA0148) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for nirsevimab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of nirsevimab. Detection was performed using HRP conjugated Human Anti-Nirsevimab Antibody, clone AbD64624pap (TZA0148P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with Fusion glycoprotein F0 of RSV at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of nirsevimab (0.15 µg/mL) plus increasing concentrations of Human Anti-Nirsevimab Antibody, clone AbD64624ad (TZA0148P) in the monovalent Fab format was added. Free nirsevimab, still capable of binding to the Fusion glycoprotein F0 of RSV coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Nirsevimab Antibody, clone AbD64624 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug nirsevimab, and its biosimilar products. It does not recognize the free Fusion glycoprotein F0 of RSV, nor nirsevimab in complex with Fusion glycoprotein F0 of RSV. This antibody can be used to measure free nirsevimab levels in serum from patients. Clone AbD64624 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0148. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0148P. A pair of anti-nirsevimab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD64624 is recommended as a detection antibody, paired with Human Anti-Nirsevimab Antibody, clone AbD64623ad (TZA0147) as the capture reagent. Nirsevimab (trade name Beyfortus) is a humanized monoclonal antibody of the IgG1/kappa isotype that targets the prefusion conformation of the RSV F protein, a glycoprotein involved in the membrane fusion step of the viral entry process and neutralises several RSV A and B strains. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli. strain.
