Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Ixekizumab Antibody, clone AbD60559ia (HCA433) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002).
A microtiter plate was coated overnight with ixekizumab, nivolumab, IgG4 isotype control, human IgG4 kappa control and human IgG4 lambda control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Ixekizumab Antibody, clone AbD60559ia (HCA433) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for ixekizumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Ixekizumab Antibody, clone AbD60559ia (HCA433, red), or clone AbD60570ia (HCA432, black) or clone AbD60555ia (HCA434, blue) or clone AbD60554ia (HCA435, green). Detection was performed using HRP conjugated ixekizumab at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL-17A at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of ixekizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Ixekizumab Antibody, clone AbD60559ia (HCA433) in the monovalent Fab format was added. Free ixekizumab, still capable of binding to the human IL-17A coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. The non-inhibitory Human Anti-Ixekizumab Antibody, clone AbD60554ia (HCA435) was used as a negative control. Data are shown as the mean of three measurements.

E Reset| Human Anti-Ixekizumab Antibody, clone AbD60559ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug ixekizumab, and its biosimilar products. It does not recognize the drug target, human Interleukin 17 (IL-17A), nor ixekizumab in complex with IL-17A. HCA433 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 0.9nM and is one of the four antibodies offered for ADA assays. Antibody HCA432 has an affinity of 0.2nM, HCA434 has an affinity of 0.6nM and HCA435 has an affinity of 0.6nM. Ixekizumab (trade name Taltz) is a humanized monoclonal antibody of the IgG4/kappa isotype that binds to interleukin 17 (IL-17A), a pro-inflammatory cytokine, and blocks its action. Blocking its effect is beneficial for use in inflammatory conditions.
