A microtiter plate was coated overnight with various antigens at a concentration of 5μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti-Isatuximab antibody, clone AbD64597ia (HCA443) at a concentration of 2μg/mL in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with isatuximab, rituximab, human IgG1 kappa control, human IgG1 lambda control and VH/VL subclass control at a concentration of 5μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti isatuximab antibody, clone AbD64597ia (HCA443) titrated to the given concentrations in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human anti isatuximab antibody, clone AbD64596ia (HCA442, red), clone AbD64597ia (HCA443, blue) or clone AbD64591ia (HCA441, black). Detection was performed using HRP conjugated isatuximab followed by QuantaBlu fluorogenic peroxidase substrate.
Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of Isatuximab (0.15 µg/mL) plus increasing concentrations of Human Anti-Isatuximab Antibody, clone AbD64597ia (HCA443) was added. Free isatuximab, still capable of binding to the human CD38, was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Isatuximab Antibody, clone AbD64597ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug isatuximab. It does not recognize the drug target, CD38, nor isatuximab in complex with CD38. This antibody can be used to measure free isatuximab levels in serum from patients. HCA443 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 0.4nM and is one of the three antibodies offered for ADA assays. Antibody HCA441 has an affinity of 0.2nM and HCA442 has an affinity of 0.2nM. Isatuximab (trade name Sarclisa) is a chimeric human/mouse IgG1/kappa, produced using recombinant DNA technology in Chinese hamster ovary (CHO) cells. Isatuximab is an IgG1-derived monoclonal antibody targeted against CD38. Its binding towards CD38 positive myeloma cells results in several downstream effects, including direct apoptosis of the affected cell and activation of immune mechanisms including antibody-dependent cell-mediated cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement dependent cyto-toxicity (CDC), all of which result in potent anti-tumour activity.
