A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Isatuximab Antibody, clone AbD64592pap (TZA0125P) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with isatuximab, rituximab, human IgG1 lambda control, human IgG4 kappa control and VH/VL subclass control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Isatuximab Antibody, clone AbD64592ad (TZA0125) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for isatuximab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of isatuximab. Detection was performed using HRP conjugated Human Anti-Isatuximab Antibody, clone AbD64592pap (TZA0125P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of isatuximab (0.15 µg/mL) plus increasing concentrations of Human Anti-Isatuximab Antibody, clone AbD64592ad (TZA0125) was added. Free isatuximab, still capable of binding to the human CD38 coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Isatuximab Antibody, clone AbD64592 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug isatuximab. It does not recognize the drug target, CD38, nor isatuximab in complex with CD38. This antibody can be used to measure free isatuximab levels in serum from patients. Clone AbD64592 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0125. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0125P. A pair of anti-isatuximab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD64592 is recommended as a detection antibody, paired with Human Anti-Isatuximab Antibody, clone AbD64599ad (TZA0124) as the capture reagent. Isatuximab (trade name Sarclisa) is a chimeric human/mouse IgG1/kappa, produced using recombinant DNA technology in Chinese hamster ovary (CHO) cells. Isatuximab is an IgG1-derived monoclonal antibody targeted against CD38. Its binding towards CD38 positive myeloma cells results in several downstream effects, including direct apoptosis of the affected cell and activation of immune mechanisms including antibody-dependent cell-mediated cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement dependent cyto-toxicity (CDC), all of which result in potent anti-tumour activity. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL phage display library and expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

- ELISA
- Clone AbD64592ad can be used in indirect ELISA to detect isatuximab. It can also be used as a detection antibody to develop a pharmacokinetic (PK) bridging ELISA for isatuximab, together with TZA0124 as the capture reagent. Note that both TZA0124 (capture) and TZA0125 (detection) contain the same peptide tags. Therefore, using anti-tag antibodies for detecting TZA0125 is not possible.
