Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti emicizumab antibody, clone AbD51683ia (HCA437) at a concentration of 2μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with emicizumab, recombinant human anti Factor IXa and anti Factor X IgG4_Pro antibodies, and nivolumab at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti emicizumab, clone AbD51683ia (HCA437) titrated to the given concentrations in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data for emicizumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human anti emicizumab antibody, clone AbD51689ia (HCA436, black) or clone AbD51683ia (HCA437, red). Detection was performed using HRP conjugated emicizumab at a concentration of 2μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.

E Reset| Human Anti-Emicizumab Antibody, clone AbD51683ia is a paratope specific, anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody emicizumab, namely the Factor X binding part of emicizumab. This antibody does not recognize free human Factor IXa or Factor X, nor emicizumab in complex with either human Factor IXa or Factor X. HCA437 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 5nM and is one of the two antibodies offered for ADA assays. Antibody HCA436 has an affinity of 1nM. Emicizumab (trade name Hemlibra) is a humanized, immunoglobulin G4 (IgG4), asymmetric bispecific monoclonal antibody. The two kappa light chains are identical. Emicizumab bridges activated factor IX (FIXa) and factor X (FX) to restore the function of missing activated factor VIII (FVIIIa) that is needed for effective haemostasis.
