A microtiter plate was coated overnight with emicizumab, recombinant human anti Factor IXa and anti factor X IgG4_Pro antibodies, and nivolumab at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti emicizumab antibody, clone AbD51689ia (HCA436) at a concentration of 2μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data for emicizumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human anti emicizumab (HCA436) titrated to the given concentrations in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data for emicizumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

E Reset| Human Anti-Emicizumab Antibody, clone AbD51689ia is a paratope specific, anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody emicizumab, namely the Factor X binding part of emicizumab. This antibody does not recognize free human Factor IXa or Factor X, nor emicizumab in complex with either human Factor IXa or Factor X. HCA436 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 1nM and is one of the two antibodies offered for ADA assays. Antibody HCA437 has an affinity of 5nM. Emicizumab (trade name Hemlibra) is a humanized, immunoglobulin G4 (IgG4), asymmetric bispecific monoclonal antibody. The two kappa light chains are identical. Emicizumab bridges activated factor IX (FIXa) and factor X (FX) to restore the function of missing activated factor VIII (FVIIIa) that is needed for effective haemostasis.
