Dupilumab antibody | AbD48333ad
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, Human Anti-Dupilumab Antibody, clone AbD48333ad (TZA016) at a concentration of 2 μg/ml was added. Detection was performed using an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, dupilumab was added at 5 µg/ml. Detection was performed using Human Anti-Dupilumab Antibody, clone AbD48333ad (TZA016) titrated to the given concentrations in PBST. An HRP conjugated anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) was added, followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of dupilumab. Detection was performed using Human Anti-Dupilumab Antibody, clone AbD48542ad (TZA017) coupled to an HRP-conjugated BiSpyCatcher2 at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with human IL-4Ra at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of dupilumab (0.15 µg/ml) plus increasing concentrations of non-inhibitory Type 2 Human Anti-Dupilumab Antibody, clone AbD48333ad (TZA016), or inhibitory Type 1 Human Anti-Dupilumab Antibody, clone AbD47092ad (TZA015), were added. Free dupilumab, still capable of binding to the human IL-4Ra coated plate, was detected using an HRP conjugated Mouse anti Human (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Dupilumab Antibody, clone AbD48333ad is a paratope specific, non-inhibitory anti-idiotypic antibody (Type 2) that specifically recognizes the free human monoclonal antibody dupilumab, and dupilumab in complex with IL-4Ra, but not free IL-4Ra. TZA016 can be used to measure total dupilumab levels (free as well as in complex with human IL-4Ra) in serum from patients. This TrailBlazer Antibody is available in an unconjugated monovalent Fab format. It may be used as capture antibody for dupilumab in a pharmacokinetic (PK) antigen detection assay together with TZA017 (AbD48542ad) coupled to an HRP-conjugated BiSpyCatcher2 as detection reagent. TZA016 can also be converted by the user into alternative antibody formats using any of the SpyCatchers available in our catalog.
