A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Brentuximab vedotin Antibody, clone AbD39655pap (TZA0146P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with Brentuximab vedotin, Brentuximab, Cetuximab, human IgG1 lambda control and human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Brentuximab vedotin Antibody, clone AbD39655pap (TZA0146P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for brentuximab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of brentuximab vedotin. Detection was performed using HRP-conjugated Human Anti-Brentuximab vedotin Antibody, clone AbD39655pap (TZA0146P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human CD30 at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of brentuximab vedotin (0.15 µg/mL) plus increasing concentrations of Human Anti-Brentuximab vedotin Antibody, clone AbD39655pap (TZA0146P) in the monovalent Fab format was added. Free brentuximab vedotin, still capable of binding to the human CD30 coated plate, was detected using HRP conjugated Mouse anti Human IgG (Fc) CH2 domain antibody (MCA647P) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Brentuximab Vedotin Antibody, clone AbD39655pap is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the monoclonal antibody drug brentuximab. It does not recognize free CD30 nor Brentuximab in complex with CD30. TZA0146P, clone AbD39655pap is a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA. A pair of anti-brentuximab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. Clone AbD39655pap is recommended as the detection antibody, paired with Human Anti-Brentuximab Vedotin Antibody in monovalent Fab format, clone AbD39649ca (HCA349) as the capture antibody. Brentuximab vedotin (Adcetris) is a chimeric mouse/human IgG1 kappa ADC, approved by the FDA for the treatment of Hodgkin’s lymphoma (HL) and systemic anaplastic large cell lymphoma (ALCL). The brentuximab antibody selectively targets tumor cells expressing the cell membrane protein CD30, and delivers the conjugated toxin, monomethyl auristatin E (MMAE). Once inside the cell the MMAE is released from the antibody by proteolytic cleavage and disrupts the microtubule network inducing cell cycle arrest and apoptosis of the malignant cells, thereby preventing tumor growth and proliferation. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
