A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Brentuximab vedotin Antibody, clone AbD39650ia (HCA350) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with brentuximab vedotin, Human Anti-CD30 Antibody (Brentuximab Biosimilar, MCA6149), cetuximab, human IgG1 kappa or human IgG1 lambda at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Brentuximab vedotin Antibody, clone AbD39650ia (HCA350) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with recombinant human CD30 at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of brentuximab vedotin (0.15 µg/ml) plus increasing concentrations of Human Anti-Brentuximab vedotin Antibody, clone AbD39650 in the monovalent Fab format was added. Free brentuximab vedotin, still capable of binding to the human CD30 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

Detection was performed using HRP conjugated brentuximab vedotin followed by QuantaBlu Fluorogenic Peroxidase Substrate. | Human Anti-Brentuximab vedotin Antibody, clone AbD39650ia is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the monoclonal antibody drug brentuximab, both alone and as the antibody drug conjugate (ADC), brentuximab vedotin. Clone AbD39650ia is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 6 nM and is one of three antibodies with different affinities offered for ADA assays. Antibody HCA351 has affinity 0.7 nM and HCA352 has affinity 1.4 nM. Brentuximab vedotin (Adcetris) is a chimeric mouse/human IgG1 kappa ADC, approved by the FDA for the treatment of Hodgkin’s lymphoma (HL) and systemic anaplastic large cell lymphoma (ALCL). The brentuximab antibody selectively targets tumor cells expressing the cell membrane protein CD30, and delivers the conjugated toxin, monomethyl auristatin E (MMAE). Once inside the cell the MMAE is released from the antibody by proteolytic cleavage and disrupts the microtubule network inducing cell cycle arrest and apoptosis of the malignant cells, thereby preventing tumor growth and proliferation.
