A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Atezolizumab Antibody, clone AbD40638ia (HCA382) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with atezolizumab, human PD-L1, human IgG1 kappa control and trastuzumab (allotype control) at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Atezolizumab Antibody, clone AbD40638ia (HCA382) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of atezolizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Atezolizumab Antibody, clone AbD40638ia (HCA382, inhibitory) or clone AbD40517 (non-inhibitory control), both in the monovalent Fab format, were added. Free atezolizumab, still capable of binding to the human PD-L1 coated plate, was detected using HRP conjugated Mouse anti Human Fc specific Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Atezolizumab Antibody, clone AbD40638ia is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the free monoclonal antibody drug atezolizumab. It does not recognize the drug target, Programmed Cell Death Ligand 1 (PD-L1), nor atezolizumab in complex with PD-L1. The antibody can be used to measure levels of free atezolizumab in patient samples. Clone AbD40638ia is a fully human recombinant monoclonal antibody with IgG1 isotype and can be used as a surrogate positive control or reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 0.4 nM, and is one of three Type 1 antibodies with different affinities offered for ADA assays. Clone AbD40638ia is also available in a monovalent Fab format with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour under product code TZA011. Atezolizumab (Tecentriq) is a human monoclonal IgG1 antibody that binds directly to Programmed Death Ligand 1 (PD-L1, CD274) and provides a dual blockade of both the PD-1 and B7.1 receptors on T-cells and antigen presenting cells. It is one of the named “checkpoint inhibitor” drugs. PD-L1/PD-1 mediated inhibition of the immune response is thereby released, including reactivation of the anti-tumour immune response without induction of antibody-dependent cellular cytotoxicity. The drug is used to treat urothelial carcinoma, non-small cell lung cancer, triple-negative breast cancer, small cell lung cancer and hepatocellular carcinoma.
