Atezolizumab antibody | AbD40638ad
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, Human Anti-Atezolizumab Antibody, clone AbD40638ad (TZA011) at a concentration of 2 μg/ml was added. Detection was performed using HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with atezolizumab, human PD-L1, human IgG1 kappa isotype control or trastuzumab (allotype control) at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Atezolizumab Antibody, clone AbD40638ad (TZA011) titrated to the given concentrations in PBST, followed by an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with human PD-L1 at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of atezolizumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Atezolizumab Antibody, clone AbD40638ad (TZA011, inhibitory) or clone AbD40517ad (non-inhibitory control) was added. Both antibodies were in the monovalent Fab fragment format. Free atezolizumab, still capable of binding to the human PD-L1 coated plate, was detected using HRP conjugated Mouse anti Human Fc specific Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of atezolizumab. Detection was performed using HRP conjugated Human Anti-Atezolizumab Antibody, clone AbD40992ia (HCA381) at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

| Human Anti-Atezolizumab Antibody, clone AbD40638ad is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the free monoclonal antibody drug atezolizumab. This is a humanized, Fc-engineered IgG1/kappa antibody with allotype nG1m1. The CH2 domain was engineered with one amino acid substitution (N297A) resulting in a non-glycosylated antibody, to impair Fcγ receptor binding and to prevent Fc-mediated depletion of cells expressing PD-L1.The antibody does not recognize free PD-L1 nor atezolizumab in complex with PD-L1. PK bridging ELISA Atezolizumab (Tecentriq) is a human monoclonal IgG1 antibody that binds directly to Programmed Death Ligand 1 (PD-L1, CD274) and provides a dual blockade of both the PD-1 and B7.1 receptors on T-cells and antigen presenting cells. It is one of the named “checkpoint inhibitor” drugs. PD-L1/PD-1 mediated inhibition of the immune response is thereby released, including reactivation of the anti-tumour immune response without induction of antibody-dependent cellular cytotoxicity. The drug is used to treat urothelial carcinoma, non-small cell lung cancer, triple-negative breast cancer, small cell lung cancer and hepatocellular carcinoma.
