Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti guselkumab antibody, clone AbD59937ad (TZA071) at a concentration of 2μg/ml followed by HRP conjugated anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with guselkumab, human IgG1 kappa control, human IgG1 lambda control, normal human serum (NHS) or human recombinant IL-23A at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti guselkumab antibody, clone AbD59937ad (TZA071) titrated to the given concentrations followed by HRP conjugated anti His tag antibody in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
Data for guselkumab are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human IL-23A at a concentration of 1μg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of guselkumab (0.15μg/ml) plus increasing concentrations of Human Anti guselkumab antibody, clone AbD59937ad (TZA071). Free guselkumab, still capable of binding to the IL-23A coated plate, was detected using HRP conjugated Mouse anti Human IgG (Fc) CH2 domain antibody (MCA647P) in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of guselkumab. Detection was performed using HRP conjugated Human anti guselkumab antibody, clone AbD59937pap (TZA071P) at a concentration of 0.5μg/mL in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.

E Reset| Human Anti-Guselkumab Antibody, clone AbD59937 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug guselkumab, and its biosimilar products. It does not recognize the drug target, the human Interleukin-23 (IL-23), nor guselkumab in complex with IL-23. This antibody can be used to measure free guselkumab levels in serum from patients. A pair of anti-guselkumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD59937 is recommended as the detection reagent, paired with Human Anti-Guselkumab Antibody, clone AbD59307ad (TZA070) as the capture antibody. TZA071 is a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog. Clone AbD59937 is also available conjugated to HRP via coupling to the BiSpyCatcher2:HRP (TZC002P) for direct detection of guselkumab. Guselkumab (trade name Tremfya) is a fully human monoclonal antibody of the IgG1/lambda isotype binding to the p19 subunit of IL-23 to block its interaction with the IL-23 receptor, subsequently inhibiting the release of pro-inflammatory cytokines and chemokines. - A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA071 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
