A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Tremelimumab Antibody, clone AbD64576ad (TZA0126) at a concentration of 2 μg/mL in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with Tremelimumab, human CTLA-4, 10% NHS, human IgG1 kappa control and human IgG1 lambda control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Tremelimumab Antibody, clone AbD64576ad (TZA0126) titrated to the given concentrations in PBST followed by an HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for tremelimumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of tremelimumab. Detection was performed using HRP conjugated Human Anti-Tremelimumab Antibody, clone AbD64580pap (TZA0127P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Tremelimumab Antibody, clone AbD64576ad is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody drug tremelimumab. It does not recognize the drug target CTLA-4, nor tremelimumab in complex with CTLA-4. This antibody can be used to measure free tremelimumab levels in serum from patients. A pair of anti-tremelimumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD64576ad is recommended as a capture antibody, paired with Human Anti-Tremelimumab Antibody, clone AbD64580pap (TZA0127P) as the detection antibody. TZA0126 is a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog. Tremelimumab (trade name Imjudo) is a fully humanized monoclonal IgG2/kappa antibody with allotype IGHG2*01. It binds to cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) to enhance T cell-mediated killing of tumours and reduces tumour growth. Tremelimumab in combination with durvalumab and platinum-based chemo-therapy is indicated for the first-line treatment of adults with metastatic non-small cell lung cancer (NSCLC) with no sensitising EGFR mutations or ALK positive mutations.
