Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti belimumab antibody, clone AbD60131ad (TZA0100) at a concentration of 2μg/ml followed by an HRP conjugated anti DYKDDDDK tag Antibody in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with belimumab, human IgG1 kappa control, human IgG1 lambda control at a concentration of 5μg/ml or 10% human normal serum (NHS). After washing and blocking with PBST + 5% BSA, detection was performed using Human anti belimumab antibody, clone AbD60131ad (TZA0100) titrated to the given concentration followed by an HRP conjugated anti His tag Antibody in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
Data for belimumab is shown as the mean of three measurements.
A microtiter plate was coated overnight with BAFF at a concentration of 1μg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of biotin conjugated belimumab (0.15μg/ml) plus increasing concentrations of Human anti belimumab antibody, clone AbD60131ad (TZA0100) in the monovalent Fab format was added. Free biotin conjugated belimumab, still capable of binding to the BAFF coated plate, was detected using HRP conjugated neutravidin followed by QuantaBlu fluorogenic peroxidase substrate. The non-inhibitory Human Anti-Belimumab Antibody, clone AbD59897ad (TZA0102) was used as a negative control.

E Reset| Human Anti-Belimumab Antibody, clone AbD60131 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug belimumab, and its biosimilar products. It does not recognize the drug target, the B-cell activating factor BAFF (CD257), nor belimumab in complex with BAFF. This antibody can be used to measure free belimumab levels in serum from patients. Clone AbD60131 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0100. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0100P. A pharmacokinetic (PK) bridging assay can be developed to measure free belimumab using clone AbD60131 in the purified Fab format TZA0100 as a capture antibody and the HRP conjugated bivalent Fab format TZA0100P for detection. Belimumab (trade name Benlysta) is a fully human monoclonal antibody of the IgG1/lambda isotype that binds to the B-cell activating factor BAFF to prevent its binding to B cells. Although part of the normal immune system, B cells are also responsible for the over-aggressive immune response observed in autoimmune diseases, such as systemic lupus erythematosus (SLE). Through its binding to BAFF, belimumab makes BAFF unavailable to B cells, which induces B cells apoptosis and therefore, a reduction in the overall number of circulating B cells. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab TZA0100 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
