A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Tremelimumab (drug/target complex) Antibody, clone AbD64586pap (TZA0128P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with CTLA-4, Tremelimumab, human IgGI kappa and lambda control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, the CTLA-4/tremelimumab complex was formed by adding 5 µg/mL tremelimumab to the wells coated with 5 µg/mL CTLA-4. Detection was performed using HRP conjugated Human Anti-Tremelimumab (Drug/Target Complex) Antibody, clone AbD64586pap (TZA0128P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate. Data for CTLA-4/tremelimumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of tremelimumab. Detection was performed using HRP conjugated Human Anti-Tremelimumab (drug/target complex) Antibody, clone AbD64586pap (TZA128P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Tremelimumab (Drug/Target Complex) Antibody, clone AbD54142 specifically recognizes the antibody drug tremelimumab when in complex with its target, cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). It does not recognize free tremelimumab nor unbound CTLA-4. The monovalent Fab format of this antibody, clone AbD64586ad (TZA0128) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA0128P for direct detection of the Tremelimumab/ CTLA-4 complex in bioanalysis assays. Clone AbD64586 can be used in pharmacokinetic (PK) antigen capture format assays to measure tremelimumab that has been captured via immobilized CTLA-4. It is ideal for bioanalytical assays to measure the level of drug or biosimilar in complex with the CTLA-4 target. Tremelimumab (trade name Imjudo) is a fully humanized monoclonal IgG2/kappa antibody with allotype IGHG2*01. It binds to cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) to enhance T cell-mediated killing of tumours and reduces tumour growth. Tremelimumab in combination with durvalumab and platinum-based chemo-therapy is indicated for the first-line treatment of adults with metastatic non-small cell lung cancer (NSCLC) with no sensitising EGFR mutations or ALK positive mutations. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

- ELISA
- Clone AbD64586ad may be used in an indirect ELISA to detect the human CTLA-4/Tremelimumab complex or to detect tremelimumab bound to immobilized human CTLA-4. This antibody can be coupled with SpyCatcher adaptors other than the BiCatcher:HRP adaptor for use as detection antibody. Note: the monovalent Fab format displays low signal in ELISA when used as detection antibody combined with an HRP labeled anti-DYKDDDDK tag antibody. Therefore, we recommend the bivalent HRP labeled format for use as detection antibody.
