Ramucirumab antibody | AbD53014ia
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Ramucirumab Antibody, clone AbD53014ia (HCA409) at a concentration of 2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with ramucirumab, VEGFR2-His, human IgG1 kappa control or NHS at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Ramucirumab Antibody, clone AbD53014ia (HCA409) titrated to the given concentrations in HISPEC diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for ramucirumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with VEGFR2-His at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of ramucirumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Ramucirumab Antibody, clone AbD53014ia (HCA409) in the monovalent Fab format was added. Free ramucirumab, still capable of binding to the VEGFR2-His coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. The non-inhibitory Human Anti-Ramucirumab Antibody, clone AbD53018ad (TZA052) was used as a negative control. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, PBST with 10% human serum was added spiked with increasing concentrations of Human Anti-Ramucirumab Antibody, clone AbD51672ia (HCA407, black), clone AbD53006ia (HCA408, blue) or clone AbD53014ia (HCA409, red). Detection was performed using HRP conjugated ramucirumab at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

| Human Anti-Ramucirumab Antibody, clone AbD53014ia is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug ramucirumab, and its biosimilar products. It does not recognize the drug target, the human vascular endothelial growth factor receptor 2 (VEGFR2), nor ramucirumab in complex with VEGFR2. This antibody can be used to measure free ramucirumab levels in serum from patients. HCA409 is a fully human recombinant monoclonal antibody with IgG1 isotype and is suitable as a surrogate positive control or calibrator in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 1.4nM and is one of the three antibodies offered for ADA assays. Antibody HCA407 has an affinity of <0.1nM and HCA408 has an affinity of 0.8nM. Ramucirumab (trade name Cyramza) is a humanized monoclonal antibody of the IgG1/kappa isotype that binds to the extracellular VEGF-binding domain of VEGFR2 to prevent VEGF ligands to bind to VEGFR2. Ramucirumab therefore blocks the activation of VEGFR2 and the downstream angiogenesis pathways involved in tumor development and progression. Ramucirumab is indicated, alone or in combination with another drug for the treatment of advanced gastric adenocarcinoma and non-small cell lung cancer. |
