A microtiter plate was coated overnight with human TNFα and infliximab at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, infliximab/TNFα complex was formed by addition of infliximab at 5 µg/ml in PBST. After washing with PBST, detection was performed using Human Anti-Infliximab (Drug/Target complex) Antibody, clone AbD18391ad (TZA009) titrated to the given concentrations in PBST followed by an HRP conjugated Mouse Anti-His tag Antibody (MCA5995P) in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for infliximab/TNFα (complex) are shown as the mean of three measurements.

| Human Anti-Infliximab (Drug/Target Complex) Antibody, clone AbD18391 specifically recognizes the antibody drug infliximab when in complex with its target, TNFα (cytokine “tumour necrosis factor-alpha”). At defined experimental levels, the antibody does not recognize free infliximab nor unbound recombinant human TNFα. However, as levels greatly increase, detection is noted. It can be used to measure levels of infliximab in patient samples. This TrailBlazer Antibody is available in two different formats, monovalent Fab unconjugated (TZA009) and bivalent Fab conjugated to HRP (TZA009P). The monovalent recombinant Fab antibody has a SpyTag incorporated, which is used to create the bivalent format by coupling two SpyTagged Fab antibodies to an HRP conjugated BiSpyCatcher2 (TZC002P). Either format can be used to develop a pharmacokinetic (PK) antigen capture assay to measure free infliximab captured via immobilized TNFα. The antibody is tagged with a SpyTag2 at the C-terminus of the Fab antibody heavy chain, enabling the user to couple it to any of the SpyCatchers available in our catalog for conversion to alternative formats in less than one hour. It has been approved for the treatment of psoriasis, Crohn’s Disease, ankylosing spondylitis, psoriatic arthritis, rheumatoid arthritis and ulcerative colitis. - A bivalent human recombinant Fab (lambda light chain) generated by coupling of the monovalent SpyTagged Fab TZA009 with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
