Size exclusion chromatography (SEC) of Human Anti-Interleukin-1β Antibody, MCA6405 (canakinumab biosimilar, research grade) was performed on an analytical HPLC instrument. Shown are monomer peaks (H2L2), calculated relative monomer portion and aggregates. Column: MAbPac SEC-1,5 μm; dimension: 4.0 x 150 mm; mobile phase: 50 mM sodium phosphate, pH 6.8, 300 mM NaCl; flow rate: 200 μl/min; inj. volume 3 μl temp.: 25 °C; detection: 280 nm; sample: mAb (1 mg/ml).
0.5 μg of Human Anti-Interleukin-1β Antibody, MCA6405 (canakinumab biosimilar, research grade) was denatured and analyzed under reducing (R) and non-reducing (NR) conditions. Bands were visualized using Coomassie staining. Full IgG (H2L2), heavy (H) and light chains (L) are indicated.
A microtiter plate was coated overnight with Human Anti-Interleukin-1β at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Interleukin-1β Antibody, MCA6405 (canakinumab biosimilar, research grade) titrated to the given concentrations in PBST followed by an HRP conjugated anti-Fab secondary antibody (STAR126P) in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti Interleukin-1β antibody, clone AbD52287ia is a research grade biosimilar of the monoclonal antibody drug canakinumab. It is a recombinant human IgG1 kappa antibody of the allotype G1m3. This product is supplied in a research grade format, i.e. in PBS buffer with preservative, it is not manufactured or formulated in the same way as the therapeutic reference product. A preservative free version is available on request. Human Anti IL-1β antibody, clone AbD52287ia, is for research use only. It is suitable as a reference standard in a pharmacokinetic (PK) bridging ELISA with anti-canakinumab antibodies. Canakinumab is a recombinant human monoclonal antibody which binds to IL-1β (interleukin-1 beta). It’s mode of action is based on the neutralization of IL-1β signaling, resulting in suppression of inflammation in patients with disorders of autoimmune origin. In June 2009, the FDA approved canakinumab for the treatment of cryopyrin-associated periodic syndromes (CAPS), a spectrum of autoinflammatory syndromes. In September 2016, the FDA approved it’s use for three additional rare and serious auto-inflammatory diseases: tumor necrosis factor receptor associated periodic syndrome (TRAPS), hyperimmunoglobulin D syndrome (HIDS)/mevalonate kinase deficiency (MKD), and familial Mediterranean fever (FMF). In June 2020, canakinumab was FDA-approved for the indication to treat active Still's disease, including adult-onset Still's disease (AOSD). This product is NOT FOR THERAPEUTIC USE. |

This product is shipped at ambient temperature. It is recommended to aliquot and store at -20°C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8°C for short term use (up to 4 weeks) and store the remaining aliquots at -20°C.
