A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Human C5 Antibody, clone AbD36700ad (TZA034) at a concentration of 2 μg/ml followed by an HRP-conjugated anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with human C5, eculizumab, human Ig4 kappa control or human IgG2 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human C5/eculizumab complex was formed by adding 5 µg/ml eculizumab to the wells coated with human C5. Detection was performed using HRP conjugated Human Anti-Human Complement C5 Antibody, clone AbD36700pap (TZA034P) titrated to the given concentrations in PBST in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for human C5 and human C5/eculizumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of human C5 (provided in 20% human serum) plus increasing concentrations of eculizumab (MCA6145) was added. Detection was performed using HRP conjugated Human Anti-Human Complement C5 Antibody, clone AbD36700pap (TZA034P) at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Human Complement C5 Antibody, clone AbD36700 specifically recognizes the human complement factor 5 (C5). The complement factor 5 (C5) forms part of the complement system and the body’s innate immune system. The complement system is vital to the body’s control of inflammation, homeostasis and defense against foreign pathogens. Downstream, C5 is cleaved into C5a and C5b by the C5 convertase. C5a increases blood vessel permeability and attracts inflammatory cells, while C5b binds with other complement components to form a membrane attack complex (MAC) to kill the pathogen. This TrailBlazer Antibody is available in two different formats: as an unconjugated monovalent Fab format (TZA034) and as a bivalent Fab antibody conjugated to HRP (TZA034P). The monovalent recombinant Fab antibody has a SpyTag incorporated, which is used to create the bivalent format by coupling two SpyTagged Fab antibodies to an HRP conjugated BiSpyCatcher2 (TZC002P).
