Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, the IL-23A/guselkumab complex was formed by adding 5μg/mL guselkumab to the wells coated with 5μg/mL IL-23A. Detection was performed using Human anti guselkumab (Drug/Target Complex) Antibody, clone AbD59967ad (TZA072) at a concentration of 2μg/ml followed by HRP conjugated anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with guselkumab, human IgG1 kappa control, human IgG1 lambda control, normal human serum (NHS) or human recombinant IL-23A at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, the IL-23A/guselkumab complex was formed by adding 5μg/mL guselkumab to the wells coated with 5μg/mL IL-23A. Detection was performed using Human anti guselkumab (drug/target complex) antibody, clone AbD59967ad (TZA072) titrated to the given concentrations followed by an HRP conjugated anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
Data for IL-23A/guselkumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of guselkumab. Detection was performed using HRP conjugated Human anti guselkumab (drug/target complex) antibody, clone AbD59967pap (TZA072P) at a concentration of 0.5μg/mL in HISPEC assay diluent (BUF049A), and QuantaBlu fluorogenic peroxidase substrate.
Data are shown as the mean of three measurements.

E Reset| Human anti guselkumab (drug/target complex) Antibody, clone AbD59967 recognizes the antibody drug guselkumab when in complex with its target, the human Interleukin-23 (IL-23). It does not recognize free guselkumab nor unbound IL-23. Clone AbD59967 can be used to develop a pharmacokinetic (PK) antigen capture assay to measure levels of guselkumab captured via immobilized IL-23. TZA072 is a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog. Clone AbD59967 is also available conjugated to HRP via coupling to the BiSpyCatcher2:HRP (TZC002P) for direct detection of the IL-23/guselkumab complex. Guselkumab (trade name Tremfya) is a fully human monoclonal antibody of the IgG1/lambda isotype binding to the p19 subunit of IL-23 to block its interaction with the IL-23 receptor, subsequently inhibiting the release of pro-inflammatory cytokines and chemokines. - A monovalent human recombinant Fab (kappa light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
