Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti emicizumab (TZA067) at a concentration of 2μg/ml in PBST followed by an HRP labelled anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A ) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with emicizumab, recombinant Human anti Factor IXa and anti Factor X IgG4_Pro antibodies, representing both Fab-arms of emicizumab, human IgG4_Pro kappa control (containing the mutation S228P in the core hinge region) and nivolumab at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti emicizumab (TZA067) titrated to the given concentrations in PBST followed by an HRP labelled anti-penta-His tag antibody (MCA5995P) in HISPEC assay diluent (BUF049A ) and QuantaBlu fluorogenic peroxidase substrate.
Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of emicizumab. Detection was performed using HRP conjugated Human anti emicizumab antibody, clone AbD50383pap (TZA067P) at a concentration of 2μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.

E Reset| Human Anti-Emicizumab Antibody, clone AbD50383 is a paratope specific, anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody emicizumab, namely the Factor X binding part of emicizumab. This antibody does not recognize free human Factor IXa or Factor X, nor emicizumab in complex with either human Factor IXa or Factor X. This antibody can be used to measure free emicizumab levels in serum from patients. TZA067 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA067. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA067P. A pair of anti-emicizumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD50383 is recommended as a detection antibody, paired with Human Anti-Emicizumab Antibody, clone AbD50384ad (TZA066) as the capture antibody. Emicizumab (trade name Hemlibra) is a humanized, immunoglobulin G4 (IgG4), asymmetric bispecific monoclonal antibody. The two kappa light chains are identical. Emicizumab bridges activated factor IX (FIXa) and factor X (FX) to restore the function of missing activated factor VIII (FVIIIa) that is needed for effective haemostasis. - A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
