Durvalumab antibody | AbD41327
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, Human Anti-Durvalumab Antibody, clone AbD41327ad (TZA004) at a concentration of 2 μg/ml was added. Detection was performed using HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with durvalumab, human PD-L1, human IgG1 kappa isotype control or human Fab VH3-23/κ3 control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Durvalumab Antibody, clone AbD41327ad (TZA004) titrated to the given concentrations in PBST, followed by an HRP conjugated anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of durvalumab. Detection was performed using HRP conjugated Human Anti-Durvalumab Antibody, clone AbD41313ia (HCA360) at a concentration of 0.5 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of durvalumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Durvalumab Antibody, clone AbD41327ad (TZA004, inhibitory) or clone AbD41238ad (non-inhibitory control) was added. Both antibodies were in the monovalent Fab fragment format. Free durvalumab, still capable of binding to the human PD-L1 coated plate, was detected using HRP conjugated Mouse anti Human Fc specific Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Durvalumab Antibody, clone AbD41327ad is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug durvalumab. The antibody does not recognize free PD-L1 nor durvalumab in complex with PD-L1. PK bridging ELISA Durvalumab (Imfinzi) is a human IgG1/kappa antibody with allotype G1m3. The CH2 domain was engineered with three amino acid substitutions (L238F, L239E, P335S) to reduce the Fc-mediated effector functions to prevent depletion of PD-L1+ T-cells. It was first approved for previously treated patients with advanced bladder cancer. It was later approved for the treatment of unresectable stage 3 NSCLC (Non-Small Cell Lung Cancer) and ES-SCLC. Durvalumab binds to PD-L1 to block the interaction with the inhibitory T cell receptor PD-1. This blockade removes the suppression of T-cells that are specific for MHC-1-presented neoepitopes on tumor cells. Removal of this inhibitory signal leads to the release of granzymes and perforins by the activated T-cells, and finally to the destruction of the tumor cells.
