A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Dupilumab Antibody, clone AbD47090ia (HCA383) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Dupilumab Antibody, clone AbD47090ia (HCA383) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of dupilumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Dupilumab Antibody, clone AbD47090ia (HCA383) or clone AbD48542ad (TZA017), both in the monovalent Fab format, were added. Free dupilumab, still capable of binding to the human IL-4Ra coated plate, was detected using HRP conjugated Mouse anti Human Fc specific antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Dupilumab Antibody, clone AbD47090ia is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody Dupilumab. HCA383 neither recognizes free IL-4Ra nor dupilumab in complex with IL-4Ra. Consequently, this antibody can be used to measure free dupilumab levels in serum from patients. Human Anti-Dupilumab Antibody, clone AbD47090ia may be used to develop and calibrate immune response assays to measure the anti-drug antibody (ADA) response in patient sera, together with HCA384. Dupilumab (Dupixent) is a human IgG4/kappa antibody inhibiting both IL‐4 and IL-13 receptors, thereby blocking these receptor signaling pathways. Dupilumab inhibits the signaling of interleukins (IL) that induce inflammatory and immunological reactions in several atopic or allergic conditions, such as eczema, allergic reaction, and rhinosinusitis.
