Daratumumab antibody | AbD44786ia
A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Daratumumab Antibody, clone AbD44786ia (HCA366) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Daratumumab Antibody, clone AbD44786ia (HCA366) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for daratumumab are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).
A microtiter plate was coated overnight with CD38 at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of daratumumab (0.15 µg/ml) plus increasing concentrations of Human Anti-Daratumumab Antibody, clone AbD44786ia (HCA366) in the monovalent Fab format was added. Free daratumumab, still capable of binding to the CD38 coated plate, was detected using Mouse Anti-Human IgG (Fc) CH2 Domain:HRP Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

Detection was performed using HRP conjugated daratumumab followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. | Human Anti-Daratumumab Antibody, clone AbD44786ia is a paratope specific, inhibitory anti-idiotypic antibody that specifically recognizes the monoclonal antibody daratumumab. It does not recognize free CD38 nor daratumumab in complex with CD38. The antibody can be used to measure free daratumumab in patient samples. Clone AbD44786ia is a fully human recombinant monoclonal antibody with IgG1 isotype and can be used as a reference standard in an anti-drug antibody (ADA) assay. This antibody has a monovalent intrinsic affinity of 9 nM, and is one of three antibodies with different affinities offered for ADA assays. Daratumumab (Darzalex) is a human IgG1/lambda antibody generated using transgenic mouse technology. It has been approved for the treatment of patients with multiple myeloma. It has orphan drug designation for multiple myeloma, diffuse large B cell lymphoma, follicular lymphoma, and mantle cell lymphoma. The antibody drug targets CD38, overexpressed by multiple myeloma cells, and indirectly induces apoptosis. In addition, daratumumab modulates CD38 enzymatic activity, inhibiting the cyclase enzyme activity and stimulating the hydrolase activity (Van de Donk et al. 2016).
