A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Belatacept Antibody, clone AbD36935pap (TZA0144P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with Belatacept, Abatacept, human IgG1 kappa control and human Fc control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Aflibercept Antibody, clone AbD36935pap (TZA0144P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for belatacept are shown as the mean of three measurements.
A microtiter plate was coated overnight with Human Anti-Belatacept Antibody, clone AbD37060 (HCA391) at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of belatacept. Detection was performed using HRP-conjugated Human Anti-Belatacept Antibody, clone AbD36935pap (TZA0144P) at a concentration of 2 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with recombinant human CD80-Fc at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of belatacept (0.15 µg/mL) plus increasing concentrations of Human Anti-Belatacept Antibody, clone AbD36935pap (TZA0144P) and AbD34644pap in the monovalent Fab format was added. Free belatacept, still capable of binding to the human CD80-Fc coated plate, was detected using QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Belatacept Antibody, clone AbD36935pap is a non-inhibitory, anti-idiotypic antibody (type 2). It recognizes the drugs belatacept and abatacept, and their biosimilar products as well as cytotoxic T-lymphocyte antigen-4 (CTLA-4), also known as CD152. The antibody binds to free belatacept, abatacept and human CTLA4 and can be used to measure free belatacept, abatacept or their biosimilars in patient samples.
TZA0144P, clone AbD36935pap is a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA. A pair of anti-belatacept antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. Clone AbD36935pap is recommended as the detection antibody, paired with Human Anti-Belatacept Antibody in monovalent Fab format, clone AbD37060 (HCA391) as the capture antibody. Belatacept (Nulojix) is a genetically engineered fusion protein which consists of the functional binding domain of modified human CTLA-4 and the Fc domain of human monoclonal immunoglobulin of the IgG1 subclass. The Fc part has the allotype IGHG1*05. Belatacept is indicated for prophylaxis of organ rejection in adult patients receiving a kidney transplant. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
