A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Aflibercept Antibody, clone AbD34644pap (TZA0141P) at a concentration of 0.2 μg/mL in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with Aflibercept, human vascular endothelial growth factor (VEGF), human VEGFR2, human VEGFR1 and human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Aflibercept Antibody, clone AbD34644pap (TZA0141P) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for aflibercept are shown as the mean of three measurements.
A microtiter plate was coated overnight with Human Anti Aflibercept Antibody, clone AbD38778da (HCA390) at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of aflibercept. Detection was performed using HRP-conjugated Human Anti-Aflibercept Antibody, clone AbD34644pap (TZA0141P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with human VEGF at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of aflibercept (0.2 µg/mL) plus increasing concentrations of Human Anti- Aflibercept Antibody, clone AbD34644pap (TZA0141P) and AbD38771pap (TZA0142P) in the monovalent Fab format was added. Free aflibercept, still capable of binding to the human VEGF coated plate, was detected using HRP conjugated Mouse anti Human IgG (Fc) CH2 domain antibody ((MCA647P) followed by QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.

E Reset| Human Anti-Aflibercept Antibody, clone AbD34644pap is a paratope specific, inhibitory antibody that specifically recognizes the free fusion protein drug aflibercept as well as the vascular endothelial growth factor receptor 1 (VEGFR1) but not the vascular endothelial growth factor receptor 2 (VEGFR2). It does not recognize free VEGF nor aflibercept in complex with VEGFR. TZA0141P, clone AbD34644pap is a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA. A pair of anti-aflibercept antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. Clone AbD34644pap is recommended as the detection antibody, paired with Human Anti-Aflibercept Antibody in monovalent Fab format, clone AbD38778da (HCA390) as the capture antibody. Aflibercept (Eylea, Zaltrap) is a recombinant fusion protein consisting of the extra-cellular domain of human VEGF receptors 1 and 2, fused to the Fc portion of human IgG1. It is used to treat the 'wet' form of age-related macular degeneration (AMD) and impaired vision due to macular oedema. It is also used in the treatment of metastatic colorectal cancer in combination with 5-fluorouracil, leucovorin and irinotecan in patients previously treated with an oxaliplatin-containing chemotherapy. - A bivalent human recombinant Fab (kappa light chain) generated by coupling of the monovalent SpyTagged Fab with a HRP conjugated BiSpyCatcher version 2 (TZC002P). The coupled antibody has two DYKDDDDK tags and three His6-tags. It contains SpyTag3 peptide (BLP086) in 5-fold molar excess to block any unreacted Catcher sites. This antibody is supplied as a liquid.
