Sandwich ELISA detection of Human HbA1c in serum samples using Human anti Human HbA1c clone AbD15792-IgG (HCA219)

Capture antibody, Mouse anti Human hemoglobin, clone 7B12 was coated at 5 μg/ml on a microtiter plate overnight at 4°C. After washing and blocking in PBST + 5% BSA, HbA1c samples (5 μg/ml total hemoglobin concentration) in PBST were added for 1hr at room temperature. Detection was performed by using Human anti HbA1C, clone AbD15783-IgG at a concentration of 2 μg/ml in PBST for 1hr at room temperature. After washing, an HRP-labeled secondary antibody, Goat anti Human IgG F(ab′)2:HRP (0500-0099) diluted 1:1000 in HISPEC Assay Diluent (BUF049A) was added. Read out was determined with QuantaBlu using a Tecan Infinite reader. Data is presented as the mean value of quadruplicate experiments

Measurement of HbA1c in patient samples using clone AbD15792-IgG

The data demonstrates the performance of HuCAL generated antibody AbD15792-IgG in the presence of abnormal hemoglobin (Hb) samples. The results show that AbD15792-IgG does not recognize HbS and HbC.

1Samples were obtained from heterozygous donors. The values reflect the percentage of abnormal Hb carried by the donor.

    • Human anti Human HbA1c antibody, clone AbD15792-IgG recognizes HbA1c, hemoglobin glycated at the N-terminal valine of the hemoglobin β chain. It does not bind to non-glycated hamoglobin. Besides binding to the normal glycated haemoglobin. Human anti Human HbA1c antibody, clone AbD15792-IgG also recognizes the glycated hemoglobin variants HbD and HbE, but does not recognise the HbS or HbC variants. Elevated HbF is not detected due to the absence of the hemoglobin β chain.

      The measurement of HbA1c levels is used to identify the average plasma glucose concentration. In diabetes mellitus, higher levels of HbA1c have been associated with cardiovascular disease and other diseases. Levels of more than 6.5% HbA1c (48 mmol/mol) are used as a criterion to diagnose diabetes. This antibody is also available in a Fab format (HCA200 ).
