* Affinity measured in the monovalent Fab format.
1 Fab-F-Spy2-H = Monovalent Fab antibody with (F) DYKDDDDK, (Spy2) SpyTag version 2, and (H) His-6-tags.
2 Fab2-FH-X22-HRP = Bivalent Fab antibody with (F) DYKDDDDK, (H) His-6-tags, and Fab-SpyTag coupled to HRP-conjugated BiCatcher.
Type 1 anti-ixekizumab antibodies inhibit the binding of the drug ixekizumab to its target, interleukin-17A (IL-17A). They are ideal for development of a pharmacokinetic (PK) bridging ELISA to measure free drug. In fully human IgG1 format, they are suitable as a surrogate positive control or calibrator for an anti-drug antibody (ADA) assay.
The Type 2 anti-ixekizumab antibodies (TZA0106, TZA0107, and TZA0107P) bind to the ixekizumab idiotope but do not inhibit the binding of ixekizumab to IL-17A. They are ideal for development of a PK bridging ELISA to measure total ixekizumab (free and IL-17A-bound ixekizumab). The Type 2 anti-ixekizumab antibody HCA435 (AbD60554ia, human IgG1 format) is an anti-idiotypic antibody (Type 2) that specifically recognizes the free, partially bound, and fully bound humanized monoclonal antibody ixekizumab. The antibody does not inhibit the binding of ixekizumab to its target IL-17A. This antibody can be used to measure free, partially bound, and fully bound ixekizumab in serum from patients in an anti-drug antibody (ADA) assay as a surrogate positive control or calibrator.
