Sarilumab antibody | AbD51309
A microtiter plate was coated over night with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Sarilumab Antibody, clone AbD51309pap (TZA047P) at a concentration of 0.5 μg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated over night with sarilumab, tocilizumab, infliximab, human IgG1 kappa control and human IgG1 lambda control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Sarilumab Antibody, clone AbD51309ad (TZA047) titrated to the given concentrations in PBST followed by a HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data is shown as the mean of three measurements.
A microtiter plate was coated over night with Human Anti-Sarilumab Antibody, clone AbD50191ad (TZA046) at a concentration of 1 µg/ml. After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of sarilumab. Detection was performed using HRP conjugated Human Anti-Sarilumab Antibody, clone AbD51309pap (TZA047P) at a concentration of 0.5 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data is shown as the mean of three measurements.
A microtiter plate was coated over night with recombinant human IL-6 receptor at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of sarilumab (0.144 µg/mL) plus increasing concentrations of Human Anti-Sarilumab Antibody, clone AbD51309ad (TZA047) in the monovalent Fab-F-Spy2-H format was added. Free sarilumab, still capable of binding to the human IL-6 receptor coated plate, was detected using a HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) and QuantaBlu fluorogenic peroxidase substrate. Data is shown as the mean of three measurements.

| Human Anti-Sarilumab Antibody, clone AbD51309 is a paratope specific, inhibitory, anti-idiotypic antibody (Type 1) that specifically recognizes the free human monoclonal antibody drug sarilumab, and its biosimilar products. It does not recognize the drug target, the human Interleukin 6 receptor (IL-6R), nor sarilumab in complex with IL-6R. This antibody can be used to measure free sarilumab levels in serum from patients. AbD51309 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA047. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA047P. A pair of anti-sarilumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD51309 is recommended as the detection antibody paired with clone AbD50191ad (TZA046) as the capture reagent. Sarilumab (trade name Kevzara) is a humanized monoclonal antibody of the IgG1/kappa isotype that suppresses the inflammatory effects of interleukin-6 (IL-6) by binding to both membrane bound and soluble IL-6 receptor forms, thus blocking cis- and trans-inflammatory signaling cascades. Through this, sarilumab blocks the IL-6 signaling pathway (measured as STAT-3 inhibition). It is indicated for the treatment of moderate to severe reactive Rheumatoid Arthritis (RA) in adult patients who are irresponsive, respond inadequately or present intolerance to disease-modifying anti-rheumatic drugs (DMARDs) or tumor necrosis factor (TNF) antagonists. It is also indicated in combination with methotrexate (MTX) or as a monotherapy when there is intolerance to MTX or when MTX administration is inappropriate. |
