A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, the CGRP/galcanezumab complex was formed by adding 5μg/mL galcanezumab to the wells coated with neutravidin and CGRP. Detection was performed using Human Anti-Galcanezumab Antibody, clone AbD64731ad (TZA0153) at a concentration of 2 μg/mL in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with galcanezumab, neutravidin, 10% normal human serum and isotype kappa control hIgG4_pro at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, CGRP was added to the wells coated with 5 µg/mL neutravidin. The CGRP/galcanezumab complex was formed by adding galcanezumab to the wells coated with 5 µg/mL neutravidin with CGRP. Detection was performed using Human Anti-Galcanezumab (Drug/Target Complex) Antibody, clone AbD64731ad (TZA0153) titrated to the given concentrations in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data for galcanezumab are shown as the mean of three measurements.

Data are shown as the mean of three measurements.

E Reset| Human Anti-Galcanezumab (Drug/Target Complex) Antibody, clone AbD64731 specifically recognizes the antibody drug galcanezumab when in complex with its target, calcitonin gene-related peptide (CGRP). It does not recognize free galcanezumab nor unbound CGRP. The monovalent Fab format of this antibody, clone AbD64731ad (TZA0153) is tagged with a SpyTag2 at the C-terminus of the Fab heavy chain, enabling the user to couple it to a SpyCatcher reagent for conversion to alternative formats in less than one hour. This antibody is also available as a bivalent Fab antibody pre-coupled to the HRP-conjugated BiSpyCatcher2 (TZC002P) under product code TZA0153P for direct detection of the Galcanezumab/ CGRP complex in bioanalysis assays. Clone AbD64731 can be used in pharmacokinetic (PK) antigen capture format assays to measure galcanezumab that has been captured via immobilized CGRP. It is ideal for bioanalytical assays to measure the level of drug or biosimilar in complex with the CGRP target. Galcanezumab (trade name Emgality) is a fully humanized recombinant monoclonal antibody of isotype IgG4 kappa. It binds to calcitonin gene-related peptide (CGRP) making it unable to bind to CGRP receptors. Galcanezumab is indicated the preventative treatment of migraines and treatment of episodic cluster headache. |

- A monovalent human recombinant Fab (lambda light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.
