A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/mL. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti- Galcaezumab Antibody, clone AbD64632ad (TZA0152) at a concentration of 2 μg/mL in PBST followed by an HRP labelled anti-DYKDDDDK tag antibody in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with galcanezumab, neutravidin, dulaglutide, human Fab VH1A/kappa1 control and human IgG4_pro kappa control at a concentration of 5 µg/mL. After washing and blocking with PBST + 5% BSA, biotinylated CGRP was added to the wells coated with 5 µg/mL neutravidin. Detection was performed using HRP conjugated Human Anti-Galcanezumab Antibody, clone AbD64632pap (TZA0152P) titrated to the given concentrations in HISPEC diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. Data for galcanezumab are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of galcanezumab. Detection was performed using HRP conjugated Human Anti-Galcanezumab Antibody, clone AbD64632pap (TZA152P) at a concentration of 2 µg/mL in HISPEC Assay Diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate. Data are shown as the mean of three measurements.
A microtiter plate was coated overnight with neutravidin at a concentration of 1 µg/mL. After washing and blocking with PBST + 5% BSA, the plate was incubated with biotinylated CGRP at a concentration of 1 µg/mL in PBST. After washing, a pre-incubated mixture of galcanezumab (0.15 µg/mL) plus increasing concentrations of Human Anti-Galcanezumab Antibody, clone AbD64632ad (TZA0152) in the monovalent Fab format was added. Free galcanezumab, still capable of binding to the CGRP-peptide coated plate was detected using HRP conjugated Mouse Anti-Human IgG (Fc) CH2 Domain Antibody (MCA647P) followed by QuantaBlu Fluorogenic Peroxidase Substrate.

E Reset| Human Anti-Galcanezumab Antibody, clone AbD64632 is a paratope specific, inhibitory anti-idiotypic antibody (Type 1) that specifically recognizes the humanized monoclonal antibody drug galcanezumab. It does not recognize the drug target calcitonin gene-related peptide (CGRP), nor galcanezumab in complex with CGRP. This antibody can be used to measure free galcanezumab levels in serum from patients. TZA0152 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0152. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0152P. A pair of anti-galcanezumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug; clone AbD64632 is recommended as a detection antibody, paired with Human Anti-Galcanezumab Antibody, clone AbD64640ad (TZA0151) as the capture antibody. Galcanezumab (trade name Emgality) is a fully humanized recombinant monoclonal antibody of isotype IgG4 kappa. It binds to calcitonin gene-related peptide (CGRP) making it unable to bind to CGRP receptors. Galcanezumab is indicated the preventative treatment of migraines and treatment of episodic cluster headache. |

- A monovalent human recombinant Fab (kappa light chain) selected from the HuCAL® phage display library, expressed in a proprietary E. coli. strain. The antibody is tagged with a DYKDDDDK tag, a SpyTag version 2 (VPTIVMVDAYKRYK) and a His6-tag (HHHHHH) at the C-terminus of the antibody heavy chain. This antibody is supplied as a liquid.

- ELISA
- Clone AbD64632ad can be used in indirect ELISA to detect galcanezumab. This antibody can be coupled with SypCatcher adaptors other than the bicatcher:HRP adaptor used in product TZA0152P (AbD64632pap) and can be used as a detection antibody to develop a pharmacokinetic (PK) bridging ELISA for galcanezumab together with TZA0151 as the capture reagent. Note that both TZA0151 (capture) and TZA0152 (detection) contain the same peptide tags. Therefore, using anti-tag antibodies for detecting TZA0152 is not possible.
