Antibody Quality and Performance Guarantee
A microtiter plate was coated overnight with various antigens at a concentration of 5μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human anti daratumumab antibody, clone AbD45015ad (TZA0104) at a concentration of 2μg/ml and an HRP conjugated anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.
A microtiter plate was coated overnight with CD38 at a concentration of 1μg/ml. After washing and blocking with PBST + 5% BSA, a pre-incubated mixture of daratumumab (0.15μg/ml) plus increasing concentrations of Human anti daratumumab antibody, clone AbD45015ad (TZA0104) was added. Free daratumumab, still capable of binding to the CD38 coated plate, was detected using HRP conjugated Mouse anti Human IgG (Fc) CH2 domain antibody, clone MK 1 A6 (MCA647P) followed by QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of daratumumab. Detection was performed using HRP conjugated Human anti daratumumab antibody, clone AbD45015pap (TZA0104P) at a concentration of 0.5μg/ml in HISPEC assay diluent (BUF049A) followed by QuantaBlu fluorogenic peroxidase substrate.

After washing and blocking with PBST + 5% BSA, detection was performed using Human anti daratumumab antibody, clone AbD45015ad (TZA0104) titrated to the given concentrations in PBST followed by an HRP conjugated anti DYKDDDDK tag antibody in HISPEC assay diluent (BUF049A) and QuantaBlu fluorogenic peroxidase substrate.

E Reset| Human anti daratumumab antibody, clone AbD45015 is a paratope specific, inhibitory anti-idiotypic antibody that recognizes the monoclonal antibody daratumumab. It does not recognize free CD38 nor daratumumab in complex with CD38. The antibody can be used to measure free daratumumab in patient samples. Human anti daratumumab antibody, clone AbD45015 is available as a monovalent Fab antibody incorporating a SpyTag at the C-terminus end of the antibody heavy chain for conversion by the end user into alternative formats using any of the SpyCatchers available in our catalog under product code TZA0104. It is also available as a bivalent Fab antibody already conjugated to HRP via coupling to the HRP conjugated BiSpyCatcher2 (TZC002P) for use in direct ELISA under product code TZA0104P. A pair of anti daratumumab antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure free drug. This antibody, in monovalent Fab format, is recommended as the capture antibody, and a bivalent format of the same clone is recommended as the detection antibody. For example, use clone AbD45015ia (HCA364) in full IgG1 format, or convert the monovalent SpyTagged Fab to a bivalent format by coupling to a SpyCatcher product. Daratumumab (trade name Darzalex) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of patients with multiple myeloma. It has orphan drug designation for multiple myeloma, diffuse large B cell lymphoma, follicular lymphoma, and mantle cell lymphoma. The antibody drug targets CD38, overexpressed by multiple myeloma cells, and indirectly induces apoptosis. In addition, daratumumab modulates CD38 enzymatic activity, inhibiting the cyclase enzyme activity and stimulating the hydrolase activity (Van de Donk et al. 2016).
