A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% BSA, detection was performed using Human Anti-Aflibercept Antibody, clone AbD38720ad (HCA389) at a concentration of 2 μg/ml followed by a HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate.
A microtiter plate was coated overnight with aflibercept, human VEGF, human VEGFR1, human VEGFR2 or human IgG1 kappa control at a concentration of 5 µg/ml. After washing and blocking with PBST + 5% BSA, the human VEGF/Aflibercept complex was formed by adding 5 µg/ml aflibercept to the wells coated with human VEGF. Detection was performed using Human Anti-Aflibercept Antibody, clone AbD38720da (HCA389) titrated to the given concentrations in PBST followed by a HRP conjugated Anti-DYKDDDDK tag Antibody in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data for aflibercept is shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% human serum was added spiked with increasing concentrations of aflibercept. Detection was performed using HRP conjugated Human Anti-Aflibercept Antibody, clone AbD38771ia (HCA388) at a concentration of 2 µg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

| Human Anti-Aflibercept Antibody, clone AbD38720da is a recombinant antibody that specifically recognizes the fusion protein drug aflibercept as well as the vascular endothelial growth factor receptor 2 (VEGFR2). It does not recognize vascular endothelial growth factor receptor 1 (VEGFR1). It does not inhibit the binding of aflibercept to its target, the vascular endothelial growth factor (VEGF); it binds to both free aflibercept and aflibercept bound to its target VEGF and can be used to measure total aflibercept and its biosimilars in bioanalytical assays. A pair of anti-aflibercept antibodies can be used to develop a pharmacokinetic (PK) bridging assay to measure total drug. This antibody, in monovalent Fab format, is recommended as the capture antibody, paired with the Human Anti-Aflibercept Antibody, clone AbD38771ia (HCA388) as the detection antibody. Aflibercept (Eylea, Zaltrap) is a recombinant fusion protein consisting of the extra-cellular domain of human VEGF receptors 1 and 2, fused to the Fc portion of human IgG1. It is used to treat the 'wet' form of age-related macular degeneration (AMD) and impaired vision due to macular oedema. It is also used in the treatment of metastatic colorectal cancer in combination with 5-fluorouracil, leucovorin and irinotecan in patients previously treated with an oxaliplatin-containing chemotherapy.
