SpyDisplay is a proprietary selection technology, harnessing SpyTag methodology. It enables a faster and more efficient selection process for discovering high-affinity antibodies.

SpyTag technology leverages an autocatalytic reaction between SpyTag, a 13-amino-acid peptide, and SpyCatcher, a 12.3 kDa protein, forming a stable isopeptide bond. SpyTag and SpyCatcher are derived from the fibronectin-binding protein of Streptococcus pyogenes. This reaction is rapid, typically occurring within minutes to an hour, and remains specific and efficient across a range of pH values (5–8), buffer conditions, and even in the presence of detergents.

The final antibodies are fully mammalian-expressed IgGs, and include a complete data package with functional characterization and sequence.

SpyDisplay uses a phagemid-based expression vector which encodes a SpyTagged Fab and a SpyCatcher-pIII integrated into the E. coli genome. In SpyDisplay, the Fab is covalently displayed via SpyTag/SpyCatcher ligation on the filamentous phage carrying the SpyCatcher fused to the pIII coat protein.
