Western blot of recombinant RB. E. Human RB protein was purified from E. coli and either Mock phosphorylated (MOCK) or phosphorylated in vitro using recombinant Cyclin D1/ CDK4 complex (D1/K4) or Cyclin E/ CDK2 complex (E/K2). The blot was probed with MCA2105 recognizing Rb phosphorylated in residue S608 (MCA2105). The blot was stripped and subsequently counter-probed with pan Rb antiserum

    • Mouse anti retinoblastoma gene protein (pSer608), clone 51B7 recognizes human and mouse Rb protein only when it has been phosphorylated at the S608 residue.

      Mouse anti retinoblastoma gene protein (pSer608), clone 51B7 does not recognize non-phosphorylated Rb or Rb phosphorylated at other sites.

      Phosphorylation of the Rb protein at S608 is dependent upon CDK4 (Zarkowska and Mittnacht 1997).

      For the detection of phosphoproteins, threonine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
