    • Rabbit anti Rat phospho-PLK1 antibody recognises the ~66 kDa serine/threonine kinase, PLK1 (polo-like kinase 1), when phosphorylated at Thr210.

      PLK1 is a member of a group of polo-like kinases which play essential roles in the progression of mitosis. PLK1 is involved in various stages of mitosis, including promotion through the activation of cyclin B1/Cdk (mitosis promoting factor), the formation of functional mitotic bipolar spindles, and the maturation of centrosomes. The timing at which PLK1 phosphorylation occurs at Ser137 and Thr210 sites, may act as a regulator of PLK1 function through the mitotic cycle.

      Antisera to PLK1 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.

      For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
