    • Rabbit anti Rat DARPP-32 antibody recognizes rat Protein phosphatase 1 regulatory subunit 1B, also known as DARPP-32 or Dopamine- and cAMP-regulated neuronal phosphoprotein. DARPP-32 is a 205 amino acid ~32 kDa member of the protein phosphatase inhibitor 1 family. Originally discovered as a substrate for cAMP-dependent protein kinase which modulates cell signalling in neurons. DARPP-32 is converted into an inhibitor of protein phosphatase-1 when phosphorylated by protein kinase A at threonine 34, as a result of dopaminergic receptor stimulation (Bibb et al. 1999). However this phosphorylation is reversed on stimulation of N-methyl-D-aspartate (NMDA) receptors by glutamate (Halpain et al. 1990). Conversely when phosphorylated at threonine-75 by CDK5, DARPP-32 becomes an inhibitor of protein kinase A (Bibb et al. 1999).

      DARPP-32 is highly expressed in striatonigral and striatopallidal projection neurons but is not observed in cholinergic and neuropeptide Y-containing striatal interneurons (Anderson and Reiner 1991). A truncated form of DARPP-32, t-DARPP is over-expressed in gastric cancer cells (El-Rifai et al. 2002) and, further t-DARPP has subsequently been identified as overexpressed in Her-2 positive breast cancer cells and implicated in resistance of these cells to Herceptin treament (Gu et al. 2009).

      DARPP-32 appears to have evolved early in vertebrate evolution and may have first appeared in gnathostomes. The immunogen sequence for Rabbit anti rat DARPP-32 antibody is identical in canine, chicken, human, mouse and Xenopus, therefore the antibody therefore is expected to recognize DARP-32 in all these species. Rabbit anti rat DARPP-32 antibody is not expected to recognize t-DARPP as this lacks the amino sequence used for immunization. For the identification of specific phospho DARPP-32 forms the use af antibodies against the threonine 34 and 75 phospho-epitopes is recommended.
