    • Rabbit anti Rat aquaporin 2 (pSer261) antibody recognizes aquaporin 2 (AQP-2), also known as ADH water channel, aquaporin-CD (AQP-CD) and collecting duct water channel protein (WCH-CD), when phosphorylated at serine 261.

      AQP2 provides the plasma membranes of the renal collecting duct with a high permeability channel to enable the movement of water across an osmotic gradient, under hormonal regulation by vasopressin. In addition to the serine 256 phosphorylation site, a novel phosphorylated serine 261 site on AQP2 exists, which decreases in expression, in response to vasopressin.

      Gene mutations in AQP2 result in hereditary nephrogenic diabetes insipidus in humans, and acquired diabetes insipidus can also occur through a decrease in the expression of the AQP2 gene, following lithium treatment. Expression of the AQP2 gene is increased during pregnancy and other conditions associated with water retention.

      For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
