    • Rabbit anti phosphorylated tryptophan hydroxylase antibody recognizes tryptophan hydroxylase (TPH), when phosphorylated at serine 58.

      TPH catalyses the 5-hydroxylation of tryptophan, in the first stage in indolamine biosynthesis (e.g. serotonin and melatonin). The activity of TPH is enhanced by phosphorylation of Ser58, by cAMP dependent protein kinase (PKA) and Ca2+/calmodulin kinase II (CAMKII).

      Antisera to phosphorylated tryptophan hydroxylase were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

      For the detection of phosphoproteins, threonine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
