A microtiter plate was coated overnight with various antigens at a concentration of 5 μg/ml. After washing and blocking with PBST + 5% milk, detection was performed using HRP conjugated Human Anti-Secukinumab (Drug/Target complex) Antibody, clone AbD36948ia (HCA374) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P).

After washing and blocking with PBST + 5% BSA, detection was performed using HRP conjugated Human Anti-Secukinumab (Drug/Target complex) Antibody, clone AbD36948ia (HCA374) titrated to the given concentrations in HISPEC Assay Diluent (BUF049A) and QuantaBlu Fluorogenic Peroxidase Substrate. HRP conjugation of the antibody used in this assay was performed using a LYNX Rapid HRP Antibody Conjugation Kit (LNK002P). Data are shown as the mean of three measurements.

After washing and blocking with PBST + 5% BSA, 10% Human Serum was added spiked with increasing concentrations of secukinumab. Detection was performed using HRP conjugated Human Anti-Secukinumab (Drug/Target complex) Antibody, clone AbD36948ia (HCA374P) at a concentration of 2 μg/ml in HISPEC Assay Diluent (BUF049A) followed by QuantaBlu Fluorogenic Peroxidase Substrate. Data are shown as the mean of three measurements.

| Human Anti-Secukinumab (Drug/Target Complex) Antibody, clone AbD36948ia specifically recognizes the human monoclonal antibody drug secukinumab when in complex with its target, interleukin 17A (IL-17A). The antibody does not recognize free secukinumab or unbound IL-17A. It can be used to measure levels of secukinumab or its biosimilars in patient samples. Clone AbD36948ia is a complex specific, non-inhibitory fully human IgG1 format antibody that can be used to develop a pharmacokinetic (PK) antigen capture assay to measure free secukinumab that has been captured via immobilized IL-17A. Protocol: PK antigen capture ELISA Secukinumab (Cosentyx) is a human IgG1/kappa antibody generated using transgenic mouse technology. It has been approved for the treatment of plaque psoriasis, ankylosing spondylitis, psoriatic arthritis and non-radiographic axial spondyloarthritis.
