The ubiquitously-expressed protein STK11 is a serine/threonine kinase, acting as a "master kinase" to activate several downstream kinases. STK11 has a downstream effect on various signalling pathways including Wnt, TGF-beta and mTOR, and is important for cell survival, polarity, and energy homeostasis. Although a significant amount is known about the downstream processes controlled by STK11, less is known about the upstream processes that activate it (Dogliotti et al. 2017). The gene encoding STK11 acts as a tumor suppressor through inhibition of the mTOR pathway, but also functions as an oncogene (Pécuchet et al. 2017). About a third of lung cancer tumors have inactive or absent STK11 protein, and STK11 deficiency in patients with lung cancer is associated with increased rates of metastasis and decreased survival (Jin et al. 2018).
