Nicastrin antibody | CD01/3H8
Nicastrin is the largest component of the gamma secetase complex, a complex that functions as a protease within the lipid bilayer to release secreted products. The gamma secretase complex is considered to be the "proteasome of the membrane" (Wolfe, 2019). Nicastrin is essential for substrate recognition by gamma secretase, and has been termed the "gatekeeper" of the complex, physically limiting access to the catalytic activity of the protease and preventing proteins from being cut inappropriately (De Strooper, 2005; Urban, 2016). Appropriate proteolysis is relevant for Alzheimer's Disease, and continuous inhibition of nicastrin in the forebrain has been shown to lead no neurodegenerative features that resemble the neuron loss on Alzheimer's Disease (Ren et al. 2019). Nicastrin is overexpressed in breast cancer and relates to worse overall survival in a subset of patients. Inhibition of nicastrin has been suggested as a therapeutic goal in breast cancer (Lombardo et al. 2012).
