Rabbit anti Human NFkB p65 antibody recognizes transcription factor p65, also known as RelA, NFKB3. Members of the NFkB family of transcription factors are critical regulators of immunity and inflammation in response to infection and injury. The most abundant form of NFkB comprises a heterodimer of the NFkB proteins p65 and p50. Under non-inflammatory conditions p65:p50 is held in the cytoplasm by inhibitor kappa b alpha (IKBa). When the cell senses an inflammatory stimulus such as tumor necrosis factor alpha (TNF alpha), IKBa is degraded and the nuclear localization signal (NLS) of p65 released. p65:p50 then enters the nucleus where it activates transcription of target genes, which include IKBa, comprising a negative feedback loop (Giridharan and Srinivasan 2018). Transient NFkB activation is important for a protective immune response, but constant activation underlies many chronic inflammatory diseases such as rheumatoid arthritis and inflammatory bowel disease (IBD) (Liu et al. 2017). NFkB is also constitutively activated in many types of cancer and has many pro-tumorigenic functions, particularly in cancers that emerge from sites of chronic inflammation (Hoesel and Schmid 2013).
